  •   繼小分子藥物和抗體藥物之后,RNA藥物正在掀起第三代藥物浪潮,應用領域由新冠病毒向其他傳染病、癌癥、罕見病以及細胞治療和基因編輯適應癥方向拓展。目前,已批準上市十余種小核酸藥物和mRNA藥物。RNA分子量大、親水性強、負電荷高,難以穿過表面同為負電荷的細胞膜,并且RNA結構極不穩(wěn)定,易被降解,所以需要遞送載體才能實現(xiàn)RNA藥物的胞內(nèi)轉染。脂質納米粒(Lipid Nanoparticle,LNP)是目前主流的非病毒遞送載體,其主要包含可電離脂質、磷脂、膽固醇和聚乙二醇化脂質四個組分。

    本課題組以新型脂質分子合成為基礎,通過大型脂質文庫構建、高通量篩選、配方優(yōu)化和活體測試并結合多種生物學手段,解析LNP“結構與遞送效率”、“結構與靶向性”和“結構與生物學效應”的關系,為滿足不同需求的RNA藥物遞送提供保障。同時,課題組開發(fā)新型RNA藥物,探究其在疫苗、蛋白替代、基因沉默、基因編輯、細胞治療等方向的生物醫(yī)學應用,為臨床轉化提供理論和技術支持。
